Анотація: This book provides the first comprehensive coverage of the quickly evolving research field of membrane contact sites (MCS). A total of 16 chapters explain their organization and role and unveil the significance of MCS for various diseases. MCS, the intracellular structures where organellar membranes come in close contact with one another, mediate the exchange of proteins, lipids, and ions. Via these functions, MCS are critical for the survival and the growth of the cell. Owing to that central role in the functioning of cells, MCS dysfunctions lead to important defects of human physiology, influence viral and bacterial infection, and cause disease such as inflammation, type II diabetes, neurodegenerative disorders, and cancer. To approach such a multifaceted topic, this volume assembles a series of chapters dealing with the full array of research about MCS and their respective roles for diseases. Most chapters also introduce the history and the state of the art of MCS research, which will initiate discussion points for the respective types of MCS for years to come. This work will appeal to all cell biologists as well as researchers on diseases that are impacted by MCS dysfunction. Additionally, it will stimulate graduate students and postdocs who will energize, drive, and develop the research field in the near future.

Анотація: This volume reviews the current state of research on immune checkpoints and offers novel concepts. It discusses the two most important immune checkpoints: T lymphocyte-associated antigen-4 (CTLA-4) and programmed cell death-1 (PD-1). It shows that antagonistic antibodies against these two molecules are highly effective in the treatment of various cancers and that PD-1 and CTLA-4 have been linked to the suppression of T-cell receptor signaling and co-stimulatory molecules. Current efforts focus on integrating knowledge gained from three interrelated fields of research: (1) understanding how the fates of different cell types are specified during development, (2) revealing the synaptic connections between identified cell types (“connectomics”) using high-resolution three-dimensional circuit anatomy, and (3) causal testing of how iden tified circuit elements contribute to visual perception and behavior.

Анотація: This book reviews the latest trends and future directions of DNA replication research. The contents reflect upon the principles that have been established through the genetic and enzymatic studies of bacterial, viral, and cellular replication during the past decades. The book begins with a historical overview of the studies on eukaryotic DNA replication by Professor Thomas Kelly, a pioneer of the field. The following chapters include genome-wide studies of replication origins and initiation factor binding, as well as the timing of DNA replications, mechanisms of initiation, DNA chain elongation and termination of DNA replication, the structural basis of functions of protein complexes responsible for execution of DNA replication, cell cycle-dependent regulation of DNA replication, the nature of replication stress and cells’ strategy to deal with the stress, and finally how all these phenomena are interconnected to genome instability and development of various diseases. Анотація: Sabine Stubler compares different proteasome isoforms and subtypes in terms of their transport and active site-related parameters applying an existing computational model. In a second step, the author extends this model to be able to describe the influence of proteasome inhibitors in in vitro experiments. The computational model, which describes the hydrolysis of short fluorogenic peptides by the 20S proteasome, is calibrated to experimental data from different proteasome isoforms using an approximate Bayesian computation approach. The dynamics of proteasome inhibitors are included into the model in order to demonstrate how to modulate the inhibitor’s transport parameters for strong or isoform-specific inhibition. Анотація: The book summarizes the emerging topic about the effects of SMF on biological samples ranging from single molecules, subcellular compartments, and cells to whole organisms, as well as the potential application of SMF in clinical treatment of cancer and other diseases. With the development and growing popularity of modern appliances, including MRI in the hospitals, the potential impact of magnetic fields on human health is invoking increasing concerns. Анотація: This thesis outlines the development of the very first technology for high-throughput analysis of paired heavy and light-chain antibody sequences, opening the door for the discovery of new antibodies and the investigation of adaptive immune responses to vaccines and diseases. By designing two new technologies for sequencing multiple mRNA transcripts from up to 10 million isolated, single cells, the author directly addresses the limitations to provide information on the identity of immune receptor pairs encoded by individual B or T lymphocytes. Previous methods for high-throughput immune repertoire sequencing have been unable to provide such information. The techniques developed in this thesis have enabled comprehensive investigation of human B-cell repertoires and have been applied for the rapid discovery of new human antibodies, to gain new insights into the development of human antibody repertoires, and for analysis of human immune responses to vaccination and disease.

Анотація: This is the second edition of a very well received book that details how the sumoylation system functions and how it modulates numerous cellular activities. SUMO is a post-translational modifier in the ubiquitin super-family that has gained recognition over the last twenty years as an essential and prevalent regulatory molecule. Individual chapters explore the biochemistry, molecular biology, and cell biology of the sumoylation system and its substrate proteins. The book is divided into three themed parts: Molecular Functions (I), Cell Growth Regulation (II), and Diseases (III). Parts I and II focus on the contribution of sumoylation to cellular activities in both the nuclear and cytoplasmic compartments. The nuclear activities covered include nucleic acid metabolism (both RNA and DNA), chromosome structure and replication, and nucleocytoplasmic transport. Анотація: In his research, David Dannheisig investigates the influence of lysine129 acetylation on the biological function of survivin including alteration of nucleocytoplasmic shuttling as well as dimerization behavior. Since survivin participates in two major hallmarks of oncogenesis, namely cell death inhibition and chromosomal segregation during the cell cycle, it reflects a valuable target in cancer therapy and research. The author establishes proximity-dependent, fluorescence-microscopic methods to quantify the interaction of survivin with the export receptor Crm1 as well as the homodimerization itself. In the future, those systems can be used to examine the feasible effect of chemical modulators which are targeting these interactions in a cellular background. The outcome achieved is an essential step towards the enhancement of potential cancer therapies.
